- Also known as: Epithalon; AEDG (Ala-Glu-Asp-Gly)
- Routes: Injection
- Sequence: AEDG (none)
- Safety tier: Strict: Avoid · Balanced: Avoid · Evidence only: Caution
- Risk axes (0 none to 3 severe): Seen harm 0, Mechanism 2, Data gap 3, Silent harm 3, Product 3
- Positive: 48% of 54 graded reports glowing or positive, shrunk toward the 49% all-report average by 8 reports' weight
- Rank: #40 of 61 research peptides

- Evidence: Small human studies — Russian, single group; plus animal
- Access: Grey market
- Onset: Sleep users mostly notice it within the first 1-3 nights; a few only near the end of a 20-day cycle or after it.
- Lasts: Taken nightly as a 10-20 day course; reported sleep benefit fades within one to a few weeks after stopping for most, some say it lasts months.
- Cost per month: ≈ €41–82 (at 0.5–1 mg/day, Semax Polska at €2.73/mg)
- Dose users report: 0.88–4.5 mg/day (median 2 mg, n=15), ≈ €72–370 a month at that dose
- Updated: 2026-10-30

**Why Avoid under the strict bar:** Its claimed mechanism, telomerase activation, is a theoretical oncogenic path, and the data come from a single group.

## What users report

In cognitive and sleep threads many report clearly deeper sleep and more REM during a cycle, sometimes persisting weeks; some get worse sleep, and a few list it among peptides that did nothing. Perhaps 30-40% notice nothing. (The general-health page covers it in more depth.)

**Effects:** deeper sleep, more REM; circadian reset; sleep through the night

**Downsides:** worse sleep in some; no effect for some; dose confusion (0.5 mg vs 5-10 mg)

**Risks:** claimed telomerase activation and overcoming of the cell-division limit (Khavinson in-vitro work): a theoretical cancer-promoting path; all efficacy data from a single research group

**Typical use:** 0.5-1 mg/night for 10-20 days, 1-2 cycles/year (community; old protocols 5-10 mg).

**Getting it:** Research-peptide vendors (lyophilised 10-50 mg vials, also N-acetyl nasal sprays); a few US clinics and compounders used to supply it before the FDA put it on its Category 2 (significant safety risk) list.

**Stacked with:** Pinealon (The usual partner, run in the same 10-20 day window for sleep; users credit Pinealon with REM and Epitalon with circadian reset.); DSIP (Common bedtime pairing for sleep; several reporters ran both, so effects are hard to separate.); SS-31 / NAD+ (Bundled into "longevity" or mitochondrial stacks taken a few times a year.)

## Risk profile

- **Seen harm 0/3:** FAERS holds no reports and no case report exists; first-hand reports describe insomnia, emotional blunting and injection lumps, none serious.
- **Mechanism 2/3:** Marketed for activating telomerase; FDA's reviewers judged it has 'the potential to be carcinogenic', and the one independent replication found it lengthened telomeres in breast-cancer cells through ALT. In vitro only, and the originating group's rodent studies found fewer or unchanged tumours.
- **Data gap 3/3:** Three human exposures ever, none reporting safety data, with no pharmacokinetic or toxicity studies; the longevity trials often cited used epithalamin, a pineal extract FDA treats as a different substance.
- **Silent harm 3/3:** Telomere-driven tumour promotion would be silent and delayed.
- **Product 3/3:** Grey-market injectable vials, plus some N-acetyl nasal sprays; US compounding resumed only under the October 2026 HHS enforcement discretion.
- **Never if:** Active cancer or a history of cancer; Pregnancy or breastfeeding (no human reproductive data)
