- Category: Sleep (Research peptide)
- Also known as: Delta sleep-inducing peptide; Emideltide
- Routes: Nasal / Injection
- Sequence: WAGGDASGE (none)
- Safety tier: Strict: Caution · Balanced: Caution · Evidence only: Caution
- Risk axes (0 none to 3 severe): Seen harm 1, Mechanism 1, Data gap 2, Silent harm 1, Product 3
- Positive: 48% of 50 graded reports glowing or positive, shrunk toward the 49% all-report average by 8 reports' weight
- Rank: #17 of 61 research peptides
- Graded reports: 50 (Glowing 8, Positive 16, Mixed 6, Nothing 11, Negative 5, Bad reaction 4)
- Evidence: Small human studies — 1980s-90s insomnia trials, mixed; a 1992 double-blind study found little effect
- Access: Grey market
- Onset: Drowsiness 15-60 min after a dose for some; sleep changes from the first night for some, after 1-3 weeks of dose-finding for others.
- Lasts: One dose covers the night; tolerance within days to weeks is common, so many use 5 nights on and 2 off, or take it as needed.
- Cost per month: ≈ €7.2–36 (at 100–500 mcg/day, Semax Polska at €2.39/mg)
- Dose users report: 200–360 mcg/day (median 250 mcg, n=6)

## What users report

Polarised. Fans report fewer night wakings, feeling rested after shorter sleep and dreams returning, and describe a narrow, person-specific 'sweet spot' dose. Roughly a quarter feel nothing, and wearable data split between more deep sleep and no change. About one in five sleep worse: early waking or paradoxical wakefulness, nightmares, rage, next-day grogginess or nausea. Tolerance within days to weeks is common.

**Effects:** fewer night-time awakenings; feels more rested; vivid/interesting dreams; relaxation after a brief head-pressure

**Downsides:** nightmares/vivid dreams; next-day grogginess; head pressure shortly after dosing; nausea (isolated); inconsistent, U-shaped dose response

**Caveat:** No known harm mechanism; old human studies reported no serious adverse events. FDA's July 2026 panel voted 6-7 against allowing it in compounding, citing missing data rather than a specific harm. Claims of insulin resistance circulate on Reddit without support.

**Typical use:** 100-500 mcg subq 30-60 min before bed; titrate slowly.

**Getting it:** Research-peptide vials and nasal sprays only; an FDA advisory panel voted 6-7 against compounding it in July 2026.

**Stacked with:** Epitalon (The most common pairing or fallback; several who felt nothing from DSIP report better sleep on Epitalon.); CJC-1295/Ipamorelin or HGH (Growth-hormone boosters taken at bedtime for deep sleep and recovery, which makes it hard to tell which compound did what.); Selank (Taken 30-60 min before DSIP to quiet racing thoughts; reports range from great sleep to nothing.)

## Risk profile

- **Seen harm 1/3:** FDA's 2026 review counted three serious events in the 107-patient IV withdrawal study (hypotension at first injection twice, once progressive on re-dosing), confounded by withdrawal itself; among about fifty first-hand reports one describes weeks of broken sleep confounded by daridorexant. Isolated and confounded.
- **Mechanism 1/3:** Its receptor is unknown; FDA noted an opioid-system mechanism never tested for toxicity or addiction liability, and the IV hypotension hints at a blood-pressure effect. Speculative rather than credible at subcutaneous or nasal doses.
- **Data gap 2/3:** About 150 people ever dosed, all IV between 1981 and 1998, in small trials (n=14-16 double-blind, 107 open-label); FDA found no subcutaneous safety data, and its advisory committee voted 6-7 against compounding it in July 2026.
- **Silent harm 1/3:** Reported harms (nightmares, insomnia, grogginess, nausea) are felt the same night or next day.
- **Product 3/3:** Reports mostly describe injecting grey-market vials; nasal sprays are the minority route.
- **Never if:** Pregnancy or breastfeeding (no human reproductive data)

## Studies

- [Schneider-Helmert 1987: DSIP for 7 nights in severe chronic insomnia](https://pubmed.ncbi.nlm.nih.gov/3622582/) (rct, n=14, 1987): In a placebo-controlled double-blind trial, night sleep and daytime alertness improved, and the gain held into the first placebo night.
- [Monti et al. 1987: short-term DSIP in chronic insomniacs (crossover)](https://pubmed.ncbi.nlm.nih.gov/3583493/) (rct, 1987): Total sleep rose only through light stage-2 sleep; deep and REM sleep were unchanged, so the authors judged it of little clinical significance.
- [Bes et al. 1992: DSIP in chronic insomnia, double-blind](https://pubmed.ncbi.nlm.nih.gov/1299794/) (rct, n=16, 1992): Sleep efficiency and latency were slightly better than placebo, but the effects were weak and subjective sleep did not change.
- [Dick et al. 1984: DSIP for alcohol and opiate withdrawal](https://pubmed.ncbi.nlm.nih.gov/6548969/) (open-label, n=107, 1984): Withdrawal symptoms resolved or improved quickly in most inpatients; a few headaches were the only side effect noted.
